研究主題

藥物基因傳遞
反轉錄病毒介導之基因傳遞 本研究室已驗證超音波駐波場可用來促進病毒轉導率。在駐波場內,懸浮細胞先被推引至壓力節點面,之後形成細胞群集並用以捕捉流轉在節點面間的病毒。藉此機制可提昇基因之傳遞率。為更進一步增加病毒轉導率,超音波設備的改良與操作參數的適化乃目前之研究方向。 非病毒型載體之研發 不同於廣在文獻上報導過的非病毒型載體,本研究室應用負離子型多醣體來研製攜帶藥物基因的奈米載體。此乃鑒於,〈一〉血漿蛋白質在攜帶藥物基因之奈米顆粒上的調理吸附作用將可能被帶負電之多醣體減低,因而使藥物基因載體較為穩定;〈二〉被細胞內噬的奈米顆粒會在內膜體中溶蝕;如此,因滲透壓差引起之內膜體漲大將可能加速藥物基因載體的逃逸率。攜帶藥物基因的多醣體奈米顆粒將經由水在油相之微乳化程序製成。多種設計的?數對奈米顆粒尺寸的影響將會做系統化的調查。完整特性化含藥物基因的多醣體奈米顆粒將會用來檢驗藥物基因傳遞之效率。
